KM0113 – LB Broth (Lennox)

Luria Bertani (LB) broth is used for the cultivation and maintenance of recombinant strains of Escherichia coli for molecular biology procedures. LB broth (Lennox) contains half of the amount of sodium chloride found in LB broth (Miller).

Enzymatic digest of casein provides the required nitrogen and carbon. Vitamin B complex required by recombinant strains of Escherichia coli are supplied by yeast extract. Sodium chloride maintains the osmotic balance and provides sodium ions for membrane transport.

Preparation

Suspend 20.0g of the medium in one litre of deionised / purified water. Allow the medium to soak whilst mixing for 10 minutes.

Sterilise at 121°C for 15 minutes with frequent stirring to completely dissolve the medium. Once cooled dispense into appropriate sterile containers.
