This has the effect of inhibiting Gram-positive organisms. The peptone acts as a nitrogen, carbon and vitamin source. Sodium chloride maintains osmotic balance. Lactose is a carbohydrate and during its fermentation causes a confined pH drop around the bacterial colony. This causes a colour change in the pH indicator, neutral red and bile precipitation.

KM0124 MacConkey Agar with Salt is a selective medium for the isolation and differentiation of bile tolerant Gram-negative (enteric) and Gram-positive (staphylococci and enterococci) organisms in all areas of bacteriology. MacConkey Agar with Salt is based on the formulation by MacConkey in 1900. Staphylococcus and Enterococcus spp. are able to grow due to the omission of crystal violet from this formulation. This medium is recommended by the World Health Organisation and the Department of Health for the bacteriological examination of water, and KM0124 is recommended as a differential primary isolation media by the UK Standards for Microbiology Investigations.

KM0031 Mannitol Salt Agar is a selective medium for the isolation of Staphylococcus aureus from clinical samples, food, cosmetics and water samples. Chapman showed that adding a higher level of sodium chloride to Mannitol Salt Agar allowed for the recovery of pathogenic staphylococci and inhibits most organisms. Coagulase positive staphylococci (e.g., S. aureus) produce yellow colonies and a surrounding yellow medium while coagulase negative staphylococci produce red colonies and no colour change of the phenol red indicator. The medium conforms to the requirements of the Harmonised EP/USP/JP, is recommended as a primary isolation media by the UK Standards for Microbiology Investigations and tested in accordance with ISO 11133:2014 and Clinical and Laboratory Standards Institute. This medium is also included in the Bacteriological Analytical Manual for cosmetics testing.

A selective medium for the isolation of Staphylococcus aureus. The high level of Sodium Chloride inhibits most other organisms and as most Staphylococcus aureus ferment Mannitol the inclusion of Phenol Red indicator gives rise to yellow colonies. Maximum Recovery Diluent (Peptone/ Saline Diluent) An osmotically controlled solution for the preparations of suspensions of food samples and for use as a diluent in dilution techniques for bacterial enumeration. The presence of a low level peptone lessens the physiological shock normally experienced by bacterial cells when they are introduced to a diluent such as Ringers Solution. The level of peptone is such that multiplication of the organisms is not possible in the time in which the sample will be present in the diluent.

Formulated to ISO 6887-1, Maximum Recovery Diluent (MRD) is a diluent designed to maintain organisms by protecting the cells from unnecessary physiological shock that may occur using other aqueous solutions. MRD is used extensively in food and environmental testing. The low level of peptone provides a protective effect but does not allow for multiplication during the short residence time in the diluent, 45 minutes. The sodium chloride prevents osmotic shock as the sample is initially diluted.

The medium is complex but includes L-Glutamic acid, Ammonium sulphate, Sodium citrate, Pyridoxine and Biotin as growth factors and Magnesium sulphate, Ferric ammonium citrate as sources of trace ions. Di-sodium phosphate and Mono-potassium phosphate are also present to maintain the pH of the medium. Further enrichment is provided by the addition of Oleic acid, Albumin and Dextrose. It should be noted that Glycerol is NOT added to this medium as Glycerol can be inhibitory for M.bovis when examining veterinary samples. This is best described as a multi-purpose medium for differentiation of enterobacteriacae that combines three individual tests into a single medium. For use the medium is inoculated by making a single stab into the medium with a straight wire (or equivalent) using a pure culture (or discrete single colony) of the test organism. Following incubation it is recommended that the medium should first of all be examined to determine whether or not the organism is motile. The presence of motility is apparent by the organism tracking out from the line of inoculation and often turning the medium turbid. Non-motile organisms generally grow within the stab line leaving the surrounding medium clear. Urease positive organisms (e.g. Proteus spp) turn the medium bright red due to the hydrolysis of the Urea in the presence of the Phenol Red Indicator often making it difficult to determine the other parameters.Indole is tested for by layering a small amount of Indole Reagent (Erlich’s or Kovac’s appear to work equally well) onto the surface of the medium and allowed a few minutes to react. A positive result is indicated by the formation of a red line at the interface of the reagent and the medium.

de Man, Rogosa & Sharpe (MRS) Agar M.R.S. Agar is intended for the cultivation and enumeration of Lactobacillus spp from a variety of sources and can be used as an alternative to Orange Serum Agar for that purpose. Magnesium Sulphate, Manganese Sulphate, Sodium Acetate and Tween are included as growth supplements. The medium can be made more specific for lactobacilli generally by lowering the pH to between 5.0 and 5.5. This has the effect of inhibiting most streptococci that may otherwise grow on the medium and can be readily confused with lactobacilli.

KM0183 Mueller Hinton Agar is used as a base to make various media for antimicrobial susceptibility testing by the disk diffusion method as described by Bauer-Kirby. First described by Mueller and Hinton, Mueller Hinton Agar has been approved as the definitive medium for antimicrobial susceptibility testing by European Committee on Antimicrobial Susceptibility Testing (EUCAST), Clinical and Laboratory Standards Institute (CLSI) and is compliant with ISO16782:2016. KM0183, when prepared as unsupplemented Mueller Hinton Agar, is suggested by EUCAST and CLSI for non-fastidious organisms such as Enterobacterales, Pseudomonas spp., Stenotrophomonas maltophilia, Acinetobacter spp., Staphylococcus spp., Enterococcus spp., Aeromonas spp., Achromobacter xylosoxidans, Vibrio spp., Bacillus spp. and Burkholderia pseudomallei. KM0183, when prepared supplemented with 5% horse blood and nicotinamide adenine dinucleotide (NAD), is suggested by EUCAST and CLSI for fastidious organisms such as Streptococcus pneumoniae, Streptococcus groups A, B, C and G, Viridans group streptococci, Haemophilus influenzae, Moraxella catarrhalis, Listeria monocytogenes, Pasteurella multocida, Campylobacter jejuni and coli, Corynebacterium spp., Aerococcus sanguinicola and urinae and Kingella kingae. Additionally, according to the UK Standards for Microbiology Investigations, when supplemented appropriately, Mueller Hinton Agar can be used in the process of identifying Haemophilus species and the HACEK group of organisms and Helicobacter species from clinical specimens. Mueller-Hinton Agar is a defined medium used primarily in Antimicrobial Sensitivity Testing using the disc diffusion technique described by Bauer-Kirby. It has been approved as the definitive medium for this purpose by the European Committee on Anitmicrobial Susceptibility Testing (EUCAST). This medium contains low levels of thymidine and thymine and controlled levels of calcium and magnesium ions. Additional supplementation of the Mueller Hinton medium using 5% Horse Blood and 20mg/L of Nicotinamide adenine dinucleotide (NAD) makes it suitable for use with the more fastidious organisms such as Streptococcus pneumoniae and Haemophilus influenzae.

Approved by the National Committee for Clinical Laboratory Standards (NCCLS) in USA this medium is approved for use in antimicrobial sensitivity testing by the disc diffusion method and is recommended particularly for use with the Bauer-Kirby Technique It is low in Thymine and Thymidine and is therefore suitable for use in testing Sulphonamides & Trimethoprim and controlled to ensure correct zone sizes with Tetracyline and Aminoglycoside antibiotics. It can be considered as an alternative to Iso-Sensitest Agar. This particular formulation has an additional 2% Sodium Chloride added to the medium making it suitable for the detection of resistance to Methicillin in staphylococci and it is included in the recommendations of British Society for Antimicrobial Chemotherapy (BSAC) for this purpose. It is not however recommended for testing of organisms requiring a CO2 enriched environment due to the pH effect on the medium. If incubation in a CO2 enriched environment is essential control organisms should be included to confirm that results have not been altered.

Once the iodine/iodide solution has been added the medium should be used immediately and cannot be stored for future use. Mycological agar is a selective medium for the isolation of pathogenic fungi, particularly dermatophytes, from clinical specimens. Enzymatic digest of soybean meal provides the required carbon, nitrogen and vitamins. Glucose is an energy source for the metabolism of fungi. The addition of chloramphenicol further reduces the risk of bacterial contamination when processing material that may be heavily contaminated with coliforms. Cycloheximide should also be added (0.4g/L) to suppress the growth of commensal yeasts and saprophytic fungi.

The medium inhibits most bacteria due to the addition of Chloramphenicol which is added to reduce the risk of bacterial contamination when processing material that may be heavily contaminated particularly with Coliforms. Cycloheximide is also added to suppress the growth of yeasts and saprophytic fungi.

Based on Fastidious Anaerobe Agar Base with added Egg Yolk Emulsion, this medium can be used to test Clostridium perfringens for phospholipase production. A zone of opalescence around the colonies is indicative of a positive reaction. It can also be used as an aid to identification of Clostridium perfringens if antitoxin is spread onto half of the plate prior to inoculation (Nagler Reaction).

BM8200 Non-acidified Glycerol Lowenstein Jensen Medium is an egg-based medium for the isolation and presumptive identification of Mycobacterium spp., particularly Mycobacterium tuberculosis. BM8200 Non-acidified Glycerol Lowenstein Jensen Medium is designed for use with sodium hydroxide (NaOH) treated specimens that have been neutralised using known buffering solutions to achieve a pH of 6.8 to 7.0 and which are inoculated directly onto the LJ medium after concentration by centrifugation. This medium will isolate most common mycobacteria including M. tuberculosis and can be used for the presumptive identification of mycobacterial infections, testing antibiotic susceptibility of isolates, and differentiating different species of Mycobacterium (by colony morphology, growth rate, biochemical characteristics, and microscopy). BM8200 is recommended by the UK Standards for Microbiology Investigations. NB: This media will not isolate wild strains of M. bovis, which requires the addition of pyruvate as a growth supplement. This is normally overcome by the inclusion of a pyruvate slope such as E&O BM8300 Non-acidified Lowenstein Jensen Pyruvate Medium.

BM8300 Non-acidified Lowenstein Jensen Pyruvate Medium is an egg-based medium for the isolation and presumptive identification of Mycobacterium spp., particularly Mycobacterium bovis. BM8300 Non-acidified Lowenstein Jensen Pyruvate Medium is designed for use with sodium hydroxide (NaOH) treated samples which have been neutralised using known buffering solutions to achieve a pH of 6.8 to 7.0 and which are inoculated directly onto LJ media after concentration by centrifugation. This medium differs from E&O BM8200 Non-acidified Glycerol Lowenstein Jensen Medium in that sodium pyruvate has replaced the glycerol, which has been demonstrated to be inhibitory to some species, particularly M. bovis. BM8300 is recommended by the UK Standards for Microbiology Investigations and tested in accordance with the principals of ISO 11133:2014. The medium contains an egg emulsion which provides the required protein and fatty acids. Sodium pyruvate is included as an alternate to glycerol to allow favourable growth of M. bovis. The coagulation of the egg albumin during preparation also provides a solid surface for inoculation purposes. Malachite green and penicillin G are incorporated into the medium to inhibit contaminating organisms other than the mycobacteria that may still be present in the specimen in the specimen after decontamination. Lowenstein-Jensen slopes should be inoculated with pre-treated specimens and incubated at 36 ± 1°C for 8 weeks, checking weekly for growth. Further incubation up to 12 weeks may be required for certain Mycobacterium species.
