  • Lowenstein-Jensen medium is based on the original formulation of Lowenstein that was subsequently modified by Jensen. It is used with fresh egg and glycerol for the isolation and presumptive identification of Mycobacterium spp., particularly Mycobacterium tuberculosis, from clinical samples. L-Asparagine and Potato starch are sources of nitrogen and vitamins in the medium. Potassium hydrogen phosphate and magnesium sulphate enhance organism growth and act as buffers. Sodium citrate and malachite green are selective agents that have inhibitory effects on organisms other than the mycobacteria. Malachite green is also incorporated into the medium to provide a colour contrast between the colonies and the medium. The required addition of egg emulsion provides the fatty acids and protein required for the metabolism of mycobacteria. Glycerol is also added which is said to enhance the growth of Mycobacterium tuberculosis however other strains of mycobacteria, particularly Mycobacterium bovis, may be inhibited by its presence. Subsequently, sodium pyruvate (12.5 g/600ml) may be used as an alternative to glycerol to encourage the growth of Mycobacterium bovis.  
  • This is a selective medium for the isolation and differentiation of bile tolerant gram-negative (enteric) and gram-positive (staphylococci and enterococci) organisms and has uses in all areas of bacteriology. It has the disadvantage that many strains of Proteus spp will spread on it and for this reason MacConkey Agar without Salt may be preferred.
  • This is a selective medium primarily for the isolation and differentiation of coliforms and non-lactose fermenting Gram –ve bacterial species from clinical, dairy, industrial and water samples. This media differs from the original MacConkey formulation with the addition of an extra 0.5% Sodium chloride, an altered Bile salt mix and the neutral red concentration was also modified. This enhanced the effect of inhibiting Gram +ve and other non-enteric bacterial species. This medium conforms to the requirements of the Harmonised USP/EP/JP.
  • This is a selective medium primarily for the isolation of Enterobacteriacae from waters & sewage. This media differs from the original MacConkey formulation in that as well as Bile Salts, Crystal Violet has been included as an additional selective agent. This has the effect of inhibiting gram-positive micrococci.
  • MacConkey agar No. 3 is a selective medium primarily for the isolation of Enterobacteriacae from waters and sewage. This media differs from the original MacConkey formulation in that as well as bile salts, crystal violet has been included as an additional selective agent. This has the effect of inhibiting Gram-positive organisms. The peptone acts as a nitrogen, carbon and vitamin source. Sodium chloride maintains osmotic balance. Lactose is a carbohydrate and during its fermentation causes a confined pH drop around the bacterial colony. This causes a colour change in the pH indicator, neutral red and bile precipitation.  
  • This is a selective medium for the isolation and differentiation of bile tolerant Gram-negative (enteric) and Gram-positive (staphylococci and enterococci) organisms and has uses in all areas of bacteriology. This medium is recommended by the World Health Organisation and the Department of Health for the bacteriological examination of water. It has the disadvantage that many strains of Proteus spp. will spread on it and for this reason MacConkey Agar without salt and crystal violet may be preferred (KM0011).  
  • Originally introduced for the isolation and differentiation of Lactose & Non-Lactose Fermenting enteric organisms the medium in this case has been modified to improve the isolation of staphylococci and enterococci. The absence of Sodium Chloride provides a low electrolyte medium that prevents spreading of most Proteus spp. Although recommended for use in the examination of urine samples in clinical laboratories it has uses in Food, Water and Dairy applications.
  • This medium was first introduced by MacConkey in 1905 for the isolation and differentiation of lactose and non-lactose fermenting enteric bacteria containing swarming strains of Proteus spp. MacConkey agar without salt and crystal violet is a differential medium that restricts swarming of Proteus spp. as sodium chloride is omitted from the medium to provide an electrolyte deficient medium. The omission of crystal violet permits the growth of Staphylococcus spp. and Enterococcus spp. The peptone is the nitrogen, carbon and vitamin source in this medium. Lactose is the fermentable carbohydrate. Lactose fermentation causes a local pH drop around the colonies which will react with the pH indicator, neutral red, and hence aids in differentiation.   • A selective medium for the enrichment of Enterobacteriaceae and the detection of Escherichia coli in pharmaceutical products. The inclusion of Bromocresol Purple indicator makes the colour change caused by acid production from the fermentation of lactose easy to read with gas formation. The presence of Ox bile helps to suppress the growth of Gram positive and non-enteric bacterial species. This formulation complies with the Harmonized USP/EP/JP.

    Malt Extract

    Malt extract is sourced from hydrolyzed vegetal protein. Due to the high concentration of carbohydrate malt extract is particularly suited for the cultivation of yeasts and moulds especially in contaminated dairy products. In microbiological culture media it provides a source of nutrients, protein, and carbon.  
  • Malt extract agar is used for the detection, isolation and enumeration yeasts and moulds. The high carbohydrate content of the medium ensures rapid growth of yeasts and moulds. Malt extract and mycological peptone provide the carbon, protein and nutrient sources required by the organisms. The acidic nature of this medium allows for optimal growth of moulds and yeasts while restricting bacterial growth. Malt extract agar can also be used for the cultivation of fungi, although with the prolonged incubation necessary, cultures may become overgrown by bacteria. The selectivity of the medium can be increased by lowering the pH to 3.5-4.0 or by the addition of selective agents such as chloramphenicol.   • This is a medium for the cultivation of yeasts and moulds. The high carbohydrate content is said to ensure rapid growth while the low pH (5.4) inhibits most bacteria. Malt Extract Agar can also be used for the cultivation of fungi although with the prolonged incubation necessary cultures may become overgrown by bacteria.

  • Mannitol Salt Agar is selective medium for the isolation of pathogenic staphylococci. The medium conforms to the requirements of the Harmonised USP/EP/JP. Chapman showed that adding a high level of sodium chloride to Phenol Red Mannitol Agar allowed for the recovery of pathogenic staphylococci. Pathogenic staphylococci produced yellow colonies whereas non-pathogenic staphylococci produced small red colonies. Pancreatic digest of casein, peptic digest of animal tissue and beef extract provide the required nitrogen, carbon and vitamins. The high level of sodium chloride inhibits most organisms other than staphylococci. Mannitol is a carbohydrate that is fermented by coagulase positive staphylococci. Phenol red is the pH indicator.
  • A selective medium for the isolation of Staphylococcus aureus. The high level of Sodium Chloride inhibits most other organisms and as most Staphylococcus aureus ferment Mannitol the inclusion of Phenol Red indicator gives rise to yellow colonies. This formulation complies with the requirements of the Harmonised USP/EP/JP. NB: Some strains of coagulase negative staphylococci can give rise to false positive results on this medium It is necessary therefore to confirm the identity of Staphylococcus aureus by other confirmatory tests (e.g. Coagulase test, Latex test etc.)
  • Selenite Mannitol Broth is an alternative to Selenite F Broth for the selective enrichment of Salmonellae spp from Clinical, Food and Environmental specimens. Based on Buffered Mannitol Broth, it is made selective by the addition of Sodium Biselenite. The fermentation of Mannitol by Salmonellae is said to correct the alkaline pH swing which can occur during incubation. Following overnight incubation subculture(s) are usually made on to one or more of the many selective enteric solid media.
  • Maximum Recovery Diluent (Peptone/ Saline Diluent) An osmotically controlled solution for the preparations of suspensions of food samples and for use as a diluent in dilution techniques for bacterial enumeration. The presence of a low level peptone lessens the physiological shock normally experienced by bacterial cells when they are introduced to a diluent such as Ringers Solution. The level of peptone is such that multiplication of the organisms is not possible in the time in which the sample will be present in the diluent.
  • Formulated to ISO 6887-1, Maximum Recovery Diluent (MRD) is a diluent designed to maintain organisms by protecting the cells from unnecessary physiological shock that may occur using other aqueous solutions. MRD is used extensively in food and environmental testing. The low level of peptone provides a protective effect but does not allow for multiplication during the short residence time in the diluent, 45 minutes. The sodium chloride prevents osmotic shock as the sample is initially diluted.

Previously known as Membrane Enriched Teepol Broth, Lauryl Sulphate has replaced Teepol 610, which is no longer available. Phenol Red is included in the medium making it possible for coliforms to be more readily detected following incubation.

The medium is also made selective by the inclusion of Ticarcillin, Polymixin B, Trimethoprim and Amphotericin B. As with egg media Glycerol is included to enhance the growth of the Mycobacteria and Malachite Green is incorporated to provide a colour contrast between the colonies and the medium as well as contributing some inhibitory effect on organisms other than mycobacteria.

The medium is complex but includes L-Glutamic acid, Ammonium sulphate, Sodium citrate, Pyridoxine and Biotin as growth factors and Magnesium sulphate, Ferric ammonium citrate as sources of trace ions. Di-sodium phosphate and Mono-potassium phosphate are also present to maintain the pH of the medium. Further enrichment is provided by the addition of Oleic acid, Albumin and Dextrose. The medium is made selective by the inclusion of Ticarcillin, Polymyxin B, Trimethoprim and Amphotericin B. Malachite green is also incorporated to provide a colour contrast between the colonies and the medium as well as contributing some inhibitory effect on organisms other than mycobacteria. It should be noted that Glycerol is NOT added to this medium as Glycerol can be inhibitory for M.bovis when examining veterinary samples. However, this product contains Lysed sheep blood, Adult bovine serum and Sodium pyruvate to enhance the growth of M.bovis.

  • Modified Campylobacter (Bolton) Selective Supplement E&O Laboratories Ltd Campylobacter Modified (Bolton) Selective Supplement (LS1054) is a supplement used to enhance the selective isolation of Campylobacter spp. primarily from clinical specimens.  
  • This is best described as a multi-purpose medium for differentiation of enterobacteriacae that combines three individual tests into a single medium. For use the medium is inoculated by making a single stab into the medium with a straight wire (or equivalent) using a pure culture (or discrete single colony) of the test organism. Following incubation it is recommended that the medium should first of all be examined to determine whether or not the organism is motile. The presence of motility is apparent by the organism tracking out from the line of inoculation and often turning the medium turbid. Non-motile organisms generally grow within the stab line leaving the surrounding medium clear. Urease positive organisms (e.g. Proteus spp) turn the medium bright red due to the hydrolysis of the Urea in the presence of the Phenol Red Indicator often making it difficult to determine the other parameters.Indole is tested for by layering a small amount of Indole Reagent (Erlich’s or Kovac’s appear to work equally well) onto the surface of the medium and allowed a few minutes to react. A positive result is indicated by the formation of a red line at the interface of the reagent and the medium.
  • de Man, Rogosa & Sharpe (MRS) Agar M.R.S. Agar is intended for the cultivation and enumeration of Lactobacillus spp from a variety of sources and can be used as an alternative to Orange Serum Agar for that purpose. Magnesium Sulphate, Manganese Sulphate, Sodium Acetate and Tween are included as growth supplements. The medium can be made more specific for lactobacilli generally by lowering the pH to between 5.0 and 5.5. This has the effect of inhibiting most streptococci that may otherwise grow on the medium and can be readily confused with lactobacilli.
  • Mueller Hinton agar is used for antimicrobial susceptibility testing by the disk diffusion method as described by Bauer-Kirby. It has been approved as the definitive medium for this purpose by the European Committee on Anitmicrobial Susceptibility Testing (EUCAST). This formulation conforms to Clinical and Laboratory Standard Institute (CLSI). Beef extract and acid hydrolysate of casein provides the required nitrogen, carbon and vitamins in the medium. Starch absorbs any toxic metabolites produced. The medium contains low levels of thymidine and thymine and controlled levels of calcium and magnesium ions. The base medium can be supplemented with blood (5 – 10%). Nicotinamide adenine dinucleotide (NAD) may be added at 20 mg/L to make the medium suitable for use with more fastidious organisms such as Streptococcus pneumoniae and Haemophilus influenzae (LS0005). It is low in Thymine and Thymidine and is therefore suitable for use in testing Sulphonamides & Trimethoprim without the addition of Lysed Blood.
  • Approved by the National Committee for Clinical Laboratory Standards (NCCLS) in USA this medium is approved for use in antimicrobial sensitivity testing by the disc diffusion method and is recommended particularly for use with the Bauer-Kirby Technique It is low in Thymine and Thymidine and is therefore suitable for use in testing Sulphonamides & Trimethoprim and controlled to ensure correct zone sizes with Tetracyline and Aminoglycoside antibiotics. It can be considered as an alternative to Iso-Sensitest Agar. This particular formulation has an additional 2% Sodium Chloride added to the medium making it suitable for the detection of resistance to Methicillin in staphylococci and it is included in the recommendations of British Society for Antimicrobial Chemotherapy (BSAC) for this purpose. It is not however recommended for testing of organisms requiring a CO2 enriched environment due to the pH effect on the medium. If incubation in a CO2 enriched environment is essential control organisms should be included to confirm that results have not been altered.

It has been approved as the definitive medium for this purpose by the European Committee on Anitmicrobial Susceptibility Testing (EUCAST). This medium contains low levels of thymidine and thymine and controlled levels of calcium and magnesium ions. Additional supplementation of the Mueller Hinton medium using 5% Horse Blood and 20mg/L of Nicotinamide adenine dinucleotide (NAD) makes it suitable for use with the more fastidious organisms such as Streptococcus pneumoniae and Haemophilus influenzae.

It is low in Thymine and Thymidine and is therefore suitable for use in testing Sulphonamides & Trimethoprim. It is sometimes used in conjunction with Mueller-Hinton Agar.

Unlike the older traditional tetrathionate broth the addition of novobiocin (40 mg/l) improves the inhibition of Proteus spp. Immediately prior to use it is necessary to add 20 ml/l of 2% iodine/iodide solution (BM0946). Once the iodine/iodide solution has been added the medium should be used immediately and cannot be stored for future use. Enzymatic digest of soybean meal provides the required carbon, nitrogen and vitamins. Glucose is an energy source for the metabolism of fungi. The addition of chloramphenicol further reduces the risk of bacterial contamination when processing material that may be heavily contaminated with coliforms. Cycloheximide should also be added (0.4g/L) to suppress the growth of commensal yeasts and saprophytic fungi.  
  • A selective medium for the isolation of fungi, particularly dermatophytes from clinical specimens, Mycological Agar is suitable for use in all areas of Mycology. The medium inhibits most bacteria due to the addition of Chloramphenicol which is added to reduce the risk of bacterial contamination when processing material that may be heavily contaminated particularly with Coliforms. Cycloheximide is also added to suppress the growth of yeasts and saprophytic fungi.

    Nagler Medium

    Based on Fastidious Anaerobe Agar Base with added Egg Yolk Emulsion, this medium can be used to test Clostridium perfringens for phospholipase production. A zone of opalescence around the colonies is indicative of a positive reaction. It can also be used as an aid to identification of Clostridium perfringens if antitoxin is spread onto half of the plate prior to inoculation (Nagler Reaction).
  • Neomycin Selective Supplement E&O Laboratories Ltd Neomycin Selective Supplement (LS0017) is an antibiotic supplement used to enhance the isolation of anaerobes from clinical specimens.  
  • This is an egg-based medium for the isolation and presumptive identification of Mycobacterium spp., particularly Mycobacterium tuberculosis. Designed for use with NaOH treated samples which are neutralised using known buffering solutions to achieve a pH of 6.8 to 7.0 and which are inoculated directly onto LJ media after concentration by centrifugation. NB: this media will NOT isolate M.bovis which requires the addition of pyruvate as a growth supplement.This is normally overcome by the inclusion of a pyruvate slope. Based on the original formulation of Lowenstein that was subsequently modified by Jensen, the medium contains glycerol and egg which provide the required protein and fatty acids. The coagulation of the egg albumin during preparation also provides a solid surface for inoculation purposes. Malachite green is incorporated into the medium to inhibit contaminating organisms other than the mycobacteria that may still be present in the specimen after decontamination. Lowenstein-Jensen slopes should be inoculated with pre-treated specimens and incubated at 35-37°C for 8 weeks in 5-10% CO2. Container caps should be left loose for the first week of incubation to allow for circulation of the carbon dioxide as this will help to stimulate growth. Caps should then be tightened to prevent any dehydration of the medium.

This medium will isolate most common mycobacteria including M.bovis. Based on the original formulation of Lowenstein that was subsequently modified by Jensen, the medium contains pyruvate and egg which provide the required protein and fatty acids. It differs from Lowenstein-Jensen Medium in that Sodium Pyruvate has replaced the Glycerol, which has been demonstrated to be inhibitory to some species, particularly M.bovis. The coagulation of the egg albumin during preparation also provides a solid surface for inoculation purposes. Malachite green is incorporated into the medium to inhibit contaminating organisms other than the mycobacteria that may still be present in the specimen after decontamination. Lowenstein-Jensen slopes should be inoculated with pre-treated specimens and incubated at 35-37°C for 8 weeks in 5-10% CO2. Container caps should be left loose for the first week of incubation to allow for circulation of the carbon dioxide as this will help to stimulate growth.
